摘要
目的:探讨大鼠急性脑缺血再灌注后Bcl-2,Bax表达及藻酸双酯钠对大鼠急性脑缺血再灌注后Bcl-2,Bax表达的影响。方法:实验于2004-01/04在华北煤炭医学院形态实验室完成。96只健康雄性Wister大鼠用随机数字表法随机分成4组:脑缺血组(42只)、藻酸双酯钠治疗组(42只)、假手术组(6只)、正常对照组(6只)。前两组又分为7组:即缺血2h再灌注1,3,6,12,24,48和72h组(n=6)。采用线栓法制作大鼠大脑中动脉阻断再灌注模型,用免疫组织化法分别检测缺血组与藻酸双酯钠治疗组大鼠脑缺血2h再灌注1,3,6,12,24,48和72hBcl-2,Bax的表达水平。结果:①皮质缺血周边区Bcl-2及Bax的阳性表达随再灌注时间不同而不同,分别于缺血2h再灌注3h犤(56.50±7.87)个/高倍视野犦及再灌注24h犤(61.00±8.88)个/高倍视野犦达高峰。②与缺血组相比,藻酸双酯钠治疗组Bax的表达于再灌注后12h犤(36.67±7.03)个/高倍视野犦、24h犤(50.50±6.09)个/高倍视野犦、48h犤(36.67±6.77)个/高倍视野犦及72h犤(29.83±5.38)个/高倍视野犦均显著减少(P<0.05,P<0.01),Bcl-2的表达未见明显变化(P>0.05)。结论:藻酸双酯钠可抑制脑缺血再灌注后Bax的表达,对神经细胞具有保护作用。
AIM: To study Bcl 2 and Bax expression following acute cerebral ischemia reperfusion in rats and the effect of alginic sodium diester(PSS) on Bcl 2 and Bax expressionin rats following acute cerebral ischemia reperfusion. METHODS: The experiment was finished in the Lab of Morphology, North China Coal Medical College from January to April 2004.Totally 96 healthy male Wister rats were divided randomly into 4 groups: cerebral ischemia group(n=42), PSS treated group(n=42), sham operation group(n=6) and control group (n=6). Cerebral ischemia group (n=42) and PSS treated group (n=42) were subdivided into 7 groups randomly: 1, 3, 6, 12 ,24 ,48 and 72 hour groups after reperfusion following two hour ischemia (n=6).The model of middle cerebral artery occlusion was made with an intraluminal filament method, and the expression of Bcl 2 and Bax were detected with immunohistochemical staining after 1 , 3 , 6 , 12 , 24 , 48 and 72 hour reperfusion following two hour cerebral ischemia in both ischemia and PSS treated groups. RESULTS: ①Positive expression of Bcl 2 and Bax was found around ischemic cortex and the level was varied to the duration of reperfusion, and reached to the peak level at 3 hour[(56.50±7.87) per visual field] and 24 hour[(61.00±8.88) per visual field]
