翻白草合剂对2型糖尿病大鼠海马神经生长因子及神经纤维蛋白含量的影响 被引量:7

Influence of herba potenillae discoloris mixture on nerve growth factors and content of neurofibrous proteins in the hippocampus of rats with type 2 diabetes mellitus

摘要 目的:探讨翻白草合剂对2型糖尿病(type2diabetesmellitus,T2DM)大鼠脑海马神经生长因子(nervegrowthfactors,NGF)、神经元纤维蛋白(neurofibrousproteins,NF含量的影响。)方法:80只Wistar大鼠随机选取10只作为正常对照组,余下的70只用高热量饲料加小剂量链脲佐菌素建立实验性T2DM大鼠模型,40只大鼠造模成功,随机分为T2DM模型组、翻白草合剂大剂量组、翻白草合剂中剂量组、翻白草合剂小剂量组,10只/组,用翻白草合剂治疗8周后,采用免疫组化法观察各组大鼠脑海马NGF、NF含量。结果:T2DM模型组大鼠脑海马NGF、NF免疫阳性细胞数目明显降低,与正常对照组比较,差异具有显著性意义(P<0.01);翻白草合剂组大鼠脑海马神经元NGF、NF免疫阳性细胞数目与T2DM模型组比较明显升高,差异具有显著性意义(P<0.01)。结论:T2DM大鼠脑海马NGF、NF含量下降,翻白草合剂可增加T2DM大鼠脑海马NGF、NF的表达。 AIM: To investigate the effects of herba potenillae discoloris (HPD) mixture on the nerve growth factors (NGF) and level of neurofibrous proteins (NF) in the hippocampus of rats with type 2 diabetes mellitus (T2DM). METHODS: Eighty Wistar rats were used in this study, and 10 of them were taken as the normal controls, and the other 70 were made into T2DM models with high calorie diet and small dose injection of streptozotocin (STZ). Forty successfully established T2DM rats were randomly divided into 4 groups with 10 rats in each group: T2DM model group, HPD large, middle and small does groups. After 8 week treatment of HPD mixture, NGF and NF contents in hippocampus of rats were determined with immunohistochemical staining in each group.RESULTS: The numbers of NGF and NF immune positive cells in the T2DM model group were significantly decreased as compared with those in the normal control group (P< 0.01); while those in the HPD mixture groups were remarkably increased as compared with those in the T2DM model group (P< 0.01). CONCLUSION: The NGF and NF contents in hippocampus of T2DM rats are decreased, and HPD mixture can increase the expressions of NGF and NF in hippocampus of T2DM rats.
