摘要
目的:检测枸杞子对遗传物质DNA受损后修复的作用,探讨枸杞子在抗衰老中的药理作用。方法:2000-03北京医科大学遗传系检测了不同浓度的枸杞培养液培养受γ射线照射的人淋巴细胞所得微核率的差异,同时在非程序DNA合成法(UDS)基础上,检测受紫外线损伤后外周血淋巴细胞诱导3H-TdR掺入细胞的量。结果:人外周血淋巴细胞微核率及微核细胞率结果:显示γ射线具有很强的诱发微核产生的作用(P<0.001);同一放射剂量下,培养液中加枸杞提取液组与不加枸杞提取液组相比,微核率及微核细胞率均极显著降低P<0.001),1g/L枸杞浓度组比0.5g/L浓度组这种抑制微(核的作用更为显著(P<0.001)。口服枸杞子后能提高正常人DNA修复能力。结论:枸杞对辐射所致DNA损伤具有良好的修复作用,为延缓细胞衰老的分子生物学研究提供理论基础。
AIM: To study the effect of medlar on DNA repair after injury, and investigate the pharmacological role of medlar against aging. METHODS:Human lymphocyte was incubated with medlar culture media of different concentrations, and then the different ratio of microkermel after treated with gamma (γ ) radiation was detected, meanwhile, different quantity of 3H TdR, which was induced by peripheral blood lymphocyte, mixed into the cell after ultraviolet damage was determined on the basis of unscheduled DNA synthesis (UDS).RESULTS: The results of human peripheral blood lymphocyte microkermel ratio and the ratio of microkermel showed that γ ray had powerful role in evoking the occurrence of microkermel (P< 0.01); With the same radiative condition, the microkermel ratio and the ratio of microkermel in the culture media with medlar were extremely and significantly decreased as compared in that without medlar (P< 0.001), and the effect was more markedly in the 1 g/L medlar group than in the 0.5 g/L group (P< 0.001). Oral medlar could enhance the ability of DNA repair after ultraviolet damage.CONCLUSION: Medlar has better effect in repairing DNA damage caused by radiation,it provides a theoretical basis for the molecular biology of postponing cellular aging.
