^(99)Tc^m-血管抑素在荷乳腺癌小鼠体内抗肿瘤血管生成的治疗效应

^(99)Tc^m-angiostatin for inhibiting neovascularization in mice bearing breast cancer

摘要 目的:血管生成是肿瘤新陈代谢过程的必要部分,血管抑素是一种强力的血管生成抑制剂,用99Tcm标记血管抑素,探讨将其用于评价肿瘤抗血管生成治疗疗效的可行性。方法:实验于2002-10/2004-10在广州军区广州总医院核医学科实验室完成。无菌操作下用氯化亚锡还原法进行血管抑素的99Tcm标记,对其产物进行标记率及放化纯测定;对荷乳腺癌EMT6瘤株的Balb/c小鼠,用不同剂量的未标记血管抑素对肿瘤局部注射,然后对小鼠尾静脉注射99Tcm-血管抑素,6h后进行平面显像,用计算机勾划感兴趣区,计算瘤/非瘤(T/NT)比值,观察显像情况与不同剂量血管抑素处理的关系。结果:99Tcm-血管抑素可使荷乳腺癌EMT6瘤株的Balb/c小鼠体内肿瘤清晰显像;经不同剂量未标记血管抑素预处理的小鼠,其T/NT比值差异具有显著性意义(F=11.178,P=0.000,ν=19),图像的T/NT比值与血管抑素的用量在统计学上呈一定程度的负相关(r=-0.920,P=0.080)。结论:99Tcm-血管抑素在荷瘤小鼠体内可浓聚于肿瘤,肿瘤对显像剂的摄取与肿瘤抗血管生成药物的应用存在一定的相关关系,99Tcm-血管抑素有可能在肿瘤抗血管生成治疗疗效评价中发挥作用,为应用于肿瘤早期诊断、治疗、预后评估奠定理论基础,为肿瘤的一级康复预防提供理论数据。 AIM:Angiogenesis is the essential process of tumor metabolism,while angiostati n(AS) is a strong inhibitor of angiogenesis.The aim of this paper is to investig ate the feasibility of angiostatin directly labeled with 99Tcm(99Tcm AS) in eva luating the effect of anti neovascularization therapy. METHODS:The experiment was conducted in the laboratory of Department of Nuclea r Medicine,Guangzhou General Hospital of Guangzhou Military Area Command of Chin ese PLA from October 2002 to October 2004. 99Tcm AS was labeled with stannous chlorde reduced approach by asepsis operation,and then the labeling efficiency a nd radiochemistry of the production were measured.Different doses of non labele d AS was locally injected into tumor of Balb/c mice bearing EMT6 tumor cell line ,then 99Tcm AS injected via tail vein and planar imaging was performed after 6 hours.The ratio of tumors to non tumors(T/NT) was counted by ROI technology and the relation between imaging and the dose of AS was observed. RESULTS:99Tcm AS could make tumor displayed clearly in Balb/c mice bearing EM T6 tumor cell line.There was significance difference in the ratio of T/NT in mic e injected with different doses of non labeled AS(F=11.178,P=0.000,ν=19).There was a negative correlation between the ratio of T/NT and the dosage of AS in st atistics to some extent(r=-0.920,P=0.080). CONCLUSION:99Tcm AS can gather strongly to tumor in tumor bearing mice.There is some certain relation between uptaking of 99Tcm AS imaging for tumor and th e application of anti angiogenesis drug.99Tcm AS is probably used for evaluati ng the effect of anti angiogenesis therapy in the treatment of tumor,which lays a theoretical foundation of early diagnosis,treatment and prognosis of tumors a nd provides theoretical data for the primary rehabilitation and intervention aft er the treatment of tumors.
