大鼠肝缺血/再灌注损伤时心肌组织能量代谢的变化 被引量:8

The changes of energy metabolism in myocardial tissue after liver ischemia/reperfusion in rats

摘要 目的:探讨大鼠肝缺血/再灌注损伤(ischemia/reperfusioninjury,I/RI)过程中心肌能量代谢的变化及其机制。方法:选健康雄性Wistar大鼠48只,建立肝脏缺血/再灌注动物模型,每组8只,共分6组。测肝组织LDH及心肌组织LDH、CPK、ATP酶活性。结果:在肝缺血/再灌注过程中,肝组织LDH在缺血组升高,与对照组比较差异有统计学意义(P<0.05);随着再灌注时间的延长,LDH活性逐渐降低,I/R2h组与I/R1h组、I/R4h组与I/R2h组比较差异均有统计学意义(P<0.05)。心肌组织LDH缺血组、I/R组与对照组比较差异无统计学意义(P>0.05),I/R1h组升高与I/R组比较差异有统计学意义(P<0.05),I/R4h组与I/R1h组比较差异有统计学意义(P<0.05)。心肌组织CPK在缺血组、I/R组、I/R1h组虽有波动,但无统计学意义(P>0.05);随着再灌注时间延长开始下降,I/R2h组与I/R1h组、I/R4h组与I/R2h组相比差异均有统计学意义(P<0.05)。心肌组织匀浆Na+-K+ATP酶和Ca2+ATP酶在I组、I/R组与对照组比较差异无统计学意义(P>0.05);I/R2h组降低,与I/R组比较差异有统计学意义(P<0.05);Na+K+ATP酶I/R4h组降低,与I/R1h组比较差异有统计学意义(P<0.05),与I/R2h组比较差异无统计学意义(P>0.05);而Ca2+ATP酶在I/R4h组降低,与I/R1h组、I/R2h组比较差异均有统计学意义(P<0.05)。 Objection: To observe the changes of myocardial tissue energy metabolism in Wistar rats, induced by liver ischemia/reperfusion injury and study its mechanism. Methods: A model of liver ischemia/reperfusion injury was established by imitation. 48 male Wistar rats were randomly divided into 6 groups as follows: the control group (the group of sham operation, CTL), simply ischemia 30 min without reperfusion (I group), reperfusion following ischemia 30 min (I/R group), 1 hour reperfusion following ischemia 30 min (I/R1 h group), 2 hours reperfusion following ischemia 30 min (I/R2 h group) and 4 hours reperfusion following ischemia 30 min (I/R4 h group). LDH in liver tissue and LDH、CPK、 Na\++-K\++ATPase、Ca^(2+)-ATPase in myocardial tissue were measured. Results: The number of LDH in liver tissue in I group is higher than that in CTL (P<0.05). But in I/R group、I/R1 h group、I/R2 h group、I/R4 h group, the number gradually decreased .In myocardial tissue , the contents of CPK、Na\++-K\++ATPase、Ca^(2+)-ATPase and LDH had not obvious changes in CTL、I group and I/R group (P>0.05), the Na\++-K\++ATPase gradually decreased in I/R1 h I group、I/R2 h group、 I/R4 h group, the Ca^(2+)-ATPase grew in I/R1 h group (P<0.05)and decreased in I/R2 h group、I/R4 h group (P<0.05). Conclusions: The results suggested that it caused the changes of myocardial tissue energetic metabolism after liver ischemia/reperfusion.
