细粒棘球蚴原头蚴体细胞的分离及其细胞形态特点 被引量:3

摘要 目的:建立体外分离细粒棘球蚴原头蚴体细胞的方法,观察其细胞形态特点。方法:分别采用0.25%胰蛋白酶消化法、玻璃匀浆器研磨法、玻璃匀浆器轻研磨与0.25%胰蛋白酶消化结合法对原头蚴体细胞进行分离。应用倒置显微镜、普通光学显微镜、透射电镜观察比较3种方法分离出的细胞形态特点。结果:3种方法均可获得原头蚴体细胞,但通过比较发现,玻璃匀浆器轻研磨法结合0.25%胰蛋白酶4℃下消化原头蚴约4~6h后所获得的细胞效果最好,其结构与形态都较完整。分离出的原头蚴体细胞在透射电镜下可见到实质细胞、单核的未分化细胞、糖原细胞和肌细胞,4种细胞具有其各自的超微结构特点。结论:初步建立了体外分离细粒棘球蚴原头蚴体细胞的方法。 Objective: To establish a way which can isolate the body cells of the protoscolices of the Echinococcus granulosus in vitro, and observe the features of cell morphology. Methods: The protoscolices were treated by 0.25% trypsin, the glass pestle, and mixed way which combined the glass pestle with 0.25% trypsin, respectively, in vitro. The features of the body cells of the protoscolices were observed under the inverted microscope, the light microscope and the transmission electron microscope. Results: Adopting these ways later, we found that the effect of the mixed way was the best, through comparing the structures of the body cells, although the cells of protoscolices can be obtained by these three ways. Combining the glass pestle with 0.25% trypsin at 4℃ to digest the protoscolices about 4~6 hours later, the structure and the morphology of the body cells of the protoscolex were relatively completely. Under the transmission electron microscope, we could see the 4 kinds of the body cells, they are parenchymatous cells, single nucleus undifferentiated cells, glycogen cells and muscle cells. These cells had their own characteristics. Conclusion: A method is iaitially established which can isolate the body cells of the protoscolices of Echinococcus granulosus in vitro.
