人β-防御素在鼻黏膜上皮细胞中的表达及意义 被引量:5

摘要 目的检测人β防御素(humanβdefensin,hBD)1,2mRNA在慢性鼻窦炎病人和正常人的鼻黏膜上皮细胞中的表达,分析hBD1和hBD2mRNA在鼻黏膜天然免疫中的作用。方法应用逆转录聚合酶链反应技术(reversetranscriptase polymerasechainreaction,RT PCR)检测16例慢性鼻窦炎病人和10例正常鼻黏膜上皮细胞中hBD1和hBD2mRNA的表达。结果hBD1mRNA在16例慢性鼻窦炎和10例正常对照鼻黏膜上皮细胞中均有表达,且两组间差异无统计学意义(P>0.05);hBD2mRNA仅表达在慢性鼻窦炎上皮细胞中,而在正常鼻黏膜上皮细胞中几乎没有表达(P<0.05)。结论鼻黏膜上皮细胞中hBD1和hBD2分别通过固有性表达和诱导性表达在鼻黏膜天然免疫中发挥重要作用。 Objective To examine the expression of humanβ-defensin-1 (hBD-1) and humanβ-defensin-2 (hBD-2) in nasal epithelial cell of normal subjects and patients with chronic sinusitis, and to investigate the role of β-defensin in the innate immunity of nasal mucosa. Methods Normal inferior turbinate mucosa was obtained from 10 subjects undergoing surgery for rhinoplasty. Inflamed nasal mucosa was taken from 16 patients undergoing endoscopic sinus surgery for chronic sunisitis. Isolated epithelial cells were used for total RNA isolation by reverse transcriptase-polymerase chain reaction (RT-PCR). Results β-defensin 1 mRNA was expressed in the all tissue samples, at levels that did not differ significantly (P>(0.05)). β-defensin 2 mRNA was detected in the nasal epithelium of patients with chronic sinusitis, but not in the epithelium of normal nasal mucosa. Its expression level was significantly higher in diseased epithelium than in normal turbinate epithelium (P<(0.05)). Conclusion These results suggest that beta-defensin 1 may play a constitutive role in nasal defenses, whereas beta-defensin 2 may be induced in response to local infection or inflammation.
