摘要
目的:建立一种快速、灵敏、准确的高效液相色谱法用于测定人血浆中盐酸二甲双胍浓度.方法:取血浆样品0.5 mL,用1.0 mL乙腈沉淀蛋白,取上清液10 μL进样,流动相为甲醇-磷酸盐(50 mmol·L-1用氢氧化钠调节pH 6.7),色谱柱为Silica柱(4.6 mm×250 mm,5 μm),紫外检测器,检测波长为233 nm.结果:盐酸二甲双胍在20~4 000 μg·L-1范围内线性关系良好,最低检测限为20μg·L-1.日内、日间精密度(RSD)小于4%.结论:该法操作简单,灵敏度高,适用于临床药动学研究.
OBJECTIVE To establish an HPLC method to determine the concentration of metformin in human plasma.METHODS 0.5mL plasma was deproteinated with acetonitrile . Metformin and phenformin (internal standard )was analyzed on hypersil silica column( 4.6 mm× 250 mm, 5 μm)and detected at 233 nm.The mobile phase was 50 mmol·L -1(NH_4)_2HPO_4(pH 6.7,adjusted with NaOH)-acetonitrile (50∶50).The flow rate was 1.0 mL·min -1.RESULTS The linear range was 20- 4 000 μg·L -1.The limit of detection was 20 μg·L -1.Within-day and between-day precision (RSD) was very good (<4%).CONCLUSION The method is simple, sensitive ,specific and reproducible.
