摘要
目的了解维甲酸(retinoicacid,RA)对ES NT3细胞的神经诱导分化的影响。方法转染NT3基因后的MESPU35胚胎干细胞(ES NT3细胞),4-/4+法诱导其神经定向分化。结果相差显微镜观察ES NT3细胞诱导分化的神经样细胞拟胚体百分率随分化时相点延长而升高;免疫细胞化学观察ES NT3细胞诱导分化形成的NF200阳性细胞和GFAP阳性细胞的比例随分化时相点延长而升高,二者总和可以达到75%。结论RA诱导ES NT3细胞神经定向分化能够产生高比例神经细胞,提示其可以作为移植治疗神经损伤的供体。
Objective To explore the effect of retinoic acid(RA) on neural differentiation of ES-NT3 embryonic stem cells.Methods ES-NT3 cells(MESPU35 cells transfected by NT3 genes of mouse)were induced by RA(4-/4+ method).Results Using RA treatment, the percentage of outgrowths of embryonic bodies containing neuron-like cells increased as the concentration of RA and days of differentiation increased, observed by phase contrast microscope. Immunocytochemistry results showed numbers of NF200 positive cells and GFAP positive cells increased after RA treatment,total number of neural cells have reached 75%.Conclusion In this study we demonstrated that differentiation of ES-NT3 cells can be induced by RA, these results suggested that ES-NT3 cells can provide plenty of multiple differentiated neural cells that can be used for transplantation.
