人嗜T淋巴细胞病毒Ⅰ型相关性脊髓病/热带痉挛性截瘫患者血清和脑脊液中细胞凋亡及凋亡相关因子的表达 被引量:4

Apoptosis and expression of Fas, TNF-α and Bcl-2 in patients’serum and cerebrospinal fluid with HAM/TSP

摘要 目的 研究人嗜T淋巴细胞病毒Ⅰ型相关性脊髓病/热带痉挛性截瘫(HAM/TSP)患者外周血淋巴细胞凋亡情况、HAM/TSP患者血清及脑脊液中sFas和sFasL表达水平及外周血Fas、肿瘤坏死引子α(TNF- α)和Bcl -2蛋白在HAM/TSP发生发展中的作用。方法 应用单细胞电泳检测HAM/TSP患者外周血淋巴细胞的凋亡情况,应用ELISA检测8例HAM/TSP患者血清和脑脊液中sFas、sFasL的水平,并用流式细胞术对患者外周血淋巴细胞上Fas、TNF -α和Bcl -2蛋白进行定量检测,并与健康对照组及其他神经系统疾病组(OIND)进行对比分析。结果 ( 1 )HAM/TSP患者外周血淋巴细胞凋亡率为13 .63%±0 .48%,与健康对照组(1 .63% ±0 .57% )比较显著增高(P<0 .05)。(2)患者血清sFas表达水平为(1 .08±0 .26)ng/ml,与OIND组[ (0 .29±0 .05)ng/ml]及健康对照组[ (0 .18±0 .07)ng/ml]比较显著增高(P<0 .05 );血清sFasL表达水平[ ( 0 .26±0 .11 )ng/ml]高于OIND组[ (0 .22±0 .04 )ng/ml]及健康对照组[ ( 0. 21±0 .05 )ng/ml],但差异无统计学意义(P>0 .05);脑脊液sFas和sFasL表达水平分别为( 0. 19±0 .07 )ng/ml和( 0. 24±0 .08 )ng/ml,显著高于OIND组[ (0 .11±0 .06)ng/ml、(0 .01±0 .01)ng/ml,P<0 .05]。(3)外周血淋巴细胞中Fas、TNF -α表达? Objective To investigate the apoptosis of peripheral blood lymphocyte cells in patients with HAM/TSP and the roles of Fas/FasL system, TNF α and Bcl 2 protein in occurrence and progression of HAM/TSP Methods The single cell gel electrophoresis (comet assay) was used to detect the apoptosis of lymphocyte cells in patients with HAM/TSP The presence of sFas, sFasL in the serum and cerebrospinal fluid (CSF) of patients with HAM/TSP was evaluated by using an enzyme linked immunosorbent assay (ELISA) The Fas, TNF α and Bcl 2 protein expressions were quanlitatively determined by flow cytometry from 8 cases of HAM/TSP, and compared with the controls and other inflammatory neurological diseases (OIND) Results (1) The apoptotic ratio of lymphocyte cells in patients with HAM/TSP (13 63%±0 48%) was significantly increased as compared to the controls (1 63%±0 57%, P <0 05) (2) Significantly increased serum levels of sFas ((1 08±0 26) ng/ml) were found in patients with HAM/TSP than in those with OIND ((0 29±0 05) ng/ml) and controls ((0 18±0 07) ng/ml, P <0 05) Serum levels of sFasL from patients with HAM/TSP ((0 26±0 11) ng/ml) showed an increasing trend as compared with controls ((0 22±0 04) ng/ml) or patients with OIND ((0 21±0 05) ng/ml, P > 0 05) Levels of sFas and sFasL in CSF from patients with HAM/TSP ((0 19±0 07) ng/ml and (0 24±0 08) ng/ml) were both significantly increased than those from OIND ((0 11±0 06) ng/ml and (0 01±0 01) ng/ml, P <0 05] (3) Patients with HAM/TSP had higher Fas and TNF α levels (180 15±105 23, 60 28±25 21) than those with OIND (41 50±21 72, 20 18±3 08) and controls (37 02±17 26, 17 60±1 50, P <0 05) Decreasing trend or significant decrease of Bcl 2 from patients with HAM/TSP (23 50±8 44) was separately shown compared with controls (27 39±8 78, P > 0 05) or patients with OIND (34 84±9 63, P <0 05) Conclusion These results suggest that the apoptosis of lymphocyte cells in patients with HAM/TSP is significantly increased, the PBMCs of the HAM/TSP patients might remain in an activated state, the patients with HAM/TSP showed have some disorder in their immune system Apoptosis of lymphocyte cells and the levels of sFas, sFasL, Fas, TNF α and Bcl 2 might play an important role in the development of HAM/TSP
