丙烯腈对体外培养鼠胚胎肢芽细胞增殖分化的研究 被引量:4

摘要 目的:研究丙烯腈(acrylonitrile,ACN)对大鼠胚胎肢芽细胞增殖分化的影响。方法:13d的大鼠胚胎脊髓组织,经取材、分离、消化后作原代细胞培养,加入不同浓度的ACN0.01,0.1,1.0,10.0,50.0,100.0,200.0μg/ml,从细胞形态学及细胞计数角度观察细胞生长与分化过程,同时测定蛋白质含量、丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性并与对照组进行比较。结果:各组ACN明显抑制胚胎肢芽细胞的增殖和分化,集落形成率明显减少,细胞体积小;其半数分化抑制剂量(ICD50)为29μg/ml,半数存活抑制剂量(ICV50)为42μg/ml,均显示剂量效应关系。结论:ACN能抑制胚胎肢芽细胞增殖和分化,可能与其能抑制蛋白质合成,并引起脂质过氧化有关。 Objective: To observe the effect of acrylonitrile (ACN) on Limb Bud Cell of rat Embryo. Methods: The primary Limb Bud Cells of rat embryo cultured 13 days were treated with ACN solution ((0.01,)0.1,1.0,10.0,50.0,100.0,200.0 μg/ml). The concentrations of protein and malondialdehyde(( MDA)) were detected , the activity of superoxide dismutase (SOD)was tested. Results: Acrylonitrile can inhibited differerntiation and proliferation and dong formation of Limb Bud cells rat embryo. The concentration of 50% inhibitions of differentiation(ICD50)was 29 μg/mL. The concentration of 50% inhibitions of cells proliferation(ICV50)was 42 μg/mL. Conclusion: The inhibition of acrylonitrile on Limb Bud cells of rat embryo may caused by inhibition of protein synthetis and inducing lipid peroxidation.[
