摘要 目的:观察Ca2+和cGMP介导的信号转导对大鼠成骨肉瘤细胞株UMR-106的C/EBPβ表达的影响。方法:以Ca2+载体A23187、cGMP类似物CPT-cGMP及CaM阻断剂KN-62等药物作用UMR106细胞株,用WesternBlotting方法检测C/EBPβ的表达。结果:A23187作用后,C/EBPβp35和C/EBPβp20的表达量在一定时间内随着作用时间的延长而增多;CaM-PK抑制剂KN-62可明显抑制A23187的这一作用;且CPTcGMP可使A23187的这一效应增强。结论:Ca2+/CaMPK和cGMP/PKG介导的信号转导能促进UMR-106细胞C/EBPβp35和C/EBPβp20的表达,并呈一定的协同作用。 Objective: To study the effects of Calcium and cGMP mediated signal transductions on the expression of C/EBPβ in rat osteosarcoma cell line UMR-106. Methods: UMR-106 cells were treated with Calcium ionophore A23187, 8-chlorophenylthio-cGMP(CPT-cGMP) and several different kinase inhibitors. The variety of the expression of C/EBPβ was detected by Western Blotting. Results: In UMR-106 cells treated with A23187, expressions of C/EBPβp35 and C/EBPβp20 were increased in a time-dependent manner, KN-62 could specifically inhibit the effect of A23187; CPT-cGMP can enhanced the expressions of C/EBPβp35 and C/EBPβp20 induced by A23187. Conclusions: Calcium/Calmodulin dependent protein kinase(Ca^(2+)/CaM-PK) and cGMP/PKG mediated signal transductions could synergistically increase the expressions of C/EBPβp35 and C/EBPβp20.
