摘要
目的 探讨wortmannin抗眼内新生血管形成的可能。方法 采用免疫荧光、流式细胞分析和Boyden小室迁移实验观察高糖视网膜Müller细胞条件培养基(HGMCM)诱导的内皮细胞增殖和迁移的影响。结果 50nmol/Lwortmannin可明显抑制HGMCM诱导的内皮细胞的增殖和迁移,使内皮细胞S期细胞百分率明显下降[ (37 .82±0 .57)%至(21. 91±0 .23)%,P<0 .01],G0 /G1 期细胞百分率显著升高[ (54 .57±1 .19)%至(65. 59±0. 41)%,P<0 .01],G2 /M期细胞增多(P<0 .05)。结论 wortmannin对HGMCM诱导的内皮细胞的增殖和迁移均有抑制作用,提示wortmannin具有抗视网膜新生血管形成的作用。
Objective To investigate the effect of wortmannin on endothelial cells prolifer ation and migration induced by high glucose Müller cell conditioned medium (HGM CM). Methods Immunofluorescence, flow cytometry and Boyden chamber migration model s were used to analyze the effect of wortmannin on endothelial cells. Results 50 nmol/L wortmannin could significantly inhibit the proliferation an d migration of endothelial cells induced by HGMCM. The percentage of endothelial cells in S phase was obviously decreased [from (37.82±0.57)% to (21.91±0 .23)%, P< 0.01], while there was an increase in the percentage of cells in G 0/G 1 phase [from (54.57±1.19)% to (65.59±0.41)%, P<0.01] and G 2/M phase (P<0.05). Conclusion Wortmannin can inhibit proliferation and migration of endothelial cel ls induced by HGMCM, suggesting that wortmannin carries an anti-angiogenetic ab ility in diseased retina.
