摘要
端粒是真核细胞中染色体末端一段特殊的DNA-蛋白质复合体,端粒酶是一种维持端粒长度的核糖核蛋白酶复合体,它能以自身RNA为模板,以3′端粒为引物合成端粒DNA并添加到染色体末端,由端粒酶RNA(telomerase RNA component,TERC)和端粒酶逆转录酶(telomerase reverse transcriptase,TERT)两个亚基组成。mterc-/-小鼠是研究端粒酶TERC亚基功能及其相关疾病的理想模型,本文就该模型在肿瘤、衰老、血液和生殖等方面的应用作一综述。
Telomeres are special DNA-protein complexes, which situated in the ends of eukaryotic chromosomes. Telomerase is a specialized ribonucleoprotein complex with reverse transcriptase activity that is capable of extending chromosome ends with specific telomeric DNA sequences by using a portion of its RNA component as a template and using 3' telomere DNA as primers. It consists of two subunits TERC and TERT. The mterc-/- mice is one of the ideal model for study the functions of subunit TERC and related diseases. This article reviewed the applications of the model on study of tumor, aging, blood and reproduction.
