非小细胞肺癌DNA含量与转移相关基因表达的相关性研究 被引量:2

Correlative studies between DNA content and metastasis associate gene expression in patient with non-small cell lung canner

摘要 目的 探讨转移相关基因CD82、CD44和Fas在肺癌中的表达与DNA含量的关系。方法 应用流式细胞仪对45例肺癌患者手术切除的新鲜癌标本中CD82、CD44、Fas蛋白及DNA含量进行检测,并与癌旁对照组(30例)和肺部良性病变组(20例)对照。结果 肺癌组CD82和Fas表达低于癌旁对照组和良性病变组,CD44表达和DI值高于癌旁组和良性病变组。CD82、CD44、Fas表达和DNA含量与肺癌的分期和分级相关。CD82和Fas表达与DI值均呈负相关(r=-0. 674,r=-0. 881,P<0. 01),CD44表达与DI值呈正相关(r=0. 542,P<0. 01),CD82和Fas表达水平与CD44蛋白的表达水平均呈负相关(r=-0. 926,r=-0. 653,P<0. 01)。结论 CD82、CD44、Fas表达及DNA含量在肺癌发生、发展和转移过程中起着不同但又相互调控的作用,联合检测这些指标对预测肿瘤的生长和转移潜能具有一定的指导意义。 Objective To observe the relationship between expression of CD_ 82、CD_ 44、Fas and DNA content in lung carcinomas.Methods The expression of CD_ 82、CD_ 44、Fas and DNA content in 45 patients with lung cancer were detected by flow-cytometry, which were compared with expression of para-cancer tissue (n=30) and 20 patients with benign lesions served as controls.Results The expression of CD_ 82 and Fas in lung cancer were lower than those in para-cancer tissue groups and benign lesion groups,while the expression of CD_ 44 and DNA index (DI) in lung cancer were higher than those in para-cancer tissue groups and benign lesion groups.The expression of CD_ 44 was positively correlated with DI (r= 0.542,P<0.01),while the expression of CD_ 82 and Fas were negatively correlated with DI in lung cancer(r=-0.674,r=-0.881,P<0.01).Conclusion CD_ 82、CD_ 44 and Fas and DNA content probably play an important regulating role in the genesis、development and metastasis of lung cancer and may be as a index to forecast potential of the tumor metastasis.
