胃癌基质金属蛋白酶7基因表达及与病理特点关系

Relationship between Matrix Metalloproteinase-7 mRNA Expression and Pathological Characteristic in Gastric Carcinoma

[目的]探讨基质金属蛋白酶7(MMP-7)基因在胃癌中的表达及与胃癌病理特点的关系。[方法]用逆转录聚合酶链式反应(RT-PCR)检测MMP-7mRNA在45例胃癌中的表达,并与相应的病理特点进行对照。[结果]随着胃癌分化程度的降低,MMP-7mRNA的表达逐渐升高(从高分化癌至未分化癌阳性率分别为57.1%、60.0%、90.9%、91.7%);随着癌细胞由黏膜层或黏膜下层向深部侵犯肌层和浆肌层时,MMP鄄7mRNA的表达阳性率大幅度提高(从黏膜层至浆膜外阳性率分别为25.0%、33.3%、84.6%、90.0%、91.7%);有淋巴管、淋巴结及静脉转移MMP鄄7mRNA表达的阳性率与无转移病例比较差异均有显著性(P<0.01)。[结论]MMP-7mRNA可作为胃癌侵袭、转移、复发的标志物,对预后评估具有重要意义。

To investigate the relationship between matrix metalloproteinase-7 mRNA expression pathological characteristic in gastric carcinoma. RT-PCR technique was used to detect the expression of MMP-7 mRNA in 45 cases with gastric carcinoma tissues and compare with the pathological characteristic. With grading increase MMP-7 mRNA expression in gastric carcinoma was getting higher. The expression rates were 57.1%, 60.0%, 90.9% and 91.7% in G1, G2, G3 and undifferentiation respectively. With cancer invading depth increase MMP-7 mRNA expression was getting higher. The expression rates were 25.0%, 33.3%, 84.6%, 90.0% and 91.7% in mucosa, submucosa, muscular and serosa invasions respectively. MMP-7 mRNA expression in patients with lymph node and vascular involvement was significantly higher than that inpatients with no involvement (P<0.01). [Conclusion] MMP-7 mRNA may be a marker of invasion, metastasis and recurrence for gastric carcinoma. It may be helpful for prognosis evaluation.
