摘要
[目的]探讨原子级或纳米级水平上揭示膀胱黏膜上皮细胞发生癌变的动态观察方法。[方法]在生理条件下,应用原子力显微镜(AFM)技术直接观测培养中的人膀胱癌BIU-87细胞,对其超微结构动态成像。[结果]得到BIU-87活细胞实时扫描图像,细胞胞膜和细胞骨架成像清晰。[结论]AFM技术可用于对膀胱癌活细胞超微结构的实时成像研究,为阐明膀胱上皮细胞发生癌变的内在机制提供了新的途径。
To observe the dynamic canceration of epthelial cell of bladder cancer at atomic or nanometer level. The ultrastructure of living BIU-87 cells of human bladder cancer cultured in one special dish was observed by using the AFM under physiological conditions and dynamic images of ultrastructure were canstructed. The real-time AFM ultrastructure images of the living BIU-87 cell were attained, and the cell membrane and the cytoskeleton were very clear. [Conclusions] The AFM technique can be used to observe and analyze the real-time ultrastructure of living cells of human bladder cancer directly. This experimental model will be a new method for elucidating the carcinogenic mechanism of the bladder normal cells at the atomic or nanometer level.
