TNF-α、IFN-γ对角细胞、成纤维细胞产生TARC、MDC的影响

摘要 目的研究TNF-α和(或)IFN-γ对皮肤角细胞、成纤维细胞产生TARC和MDC的影响。方法采用ELISA、RT-PCR等方法检测人角细胞株-HaCaT细胞和人成纤维细胞株-NG1RGB细胞TARC、MDC的表达。结果TNF-α和(或)IFN-γ作用下的皮肤角细胞产生TARC,而成纤维细胞不能表达;MDC的表达情况与TARC的结果相同。结论在TNF-α和(或)IFN-γ作用的角细胞、成纤维细胞上,TARC和MDC两种趋化因子具有相同的产生机制。 Objective In order to compare the differential regulation of TARC and MDC production by tumor necrosis factor-(TNF-α) and/or interferon(IFN-γ) in keratinocytes and fibroblasts. Methods Cell culture (HaCaT cells, a human keratinocyte cell line, and NG1RGB cells, a human skin fibroblast cell line),ELISA and RT-PCR were used. Results It was keratinocytes, not fibroblasts, which expressed TARC upon induction by TNF-α and IFN-γ The result of MDC is the same as TARC. Conclusions The mechanism of TARC and MDC production was similar by TNF-α and/or IFN-γ in human keratinocytes and fibrolasts.
