淋巴因子激活的骨髓细胞对混合淋巴细胞培养的抑制作用 被引量:1

摘要 采用单向混合淋巴细胞培养(MLC)方法,从不同主要组织相容性抗原(H-2)单型小鼠的多种组合中,验证淋巴因子激话的骨髓细胞(LABMC)的Veto效应。结果表明:LABMC于培养当天加入单向MLC,在与反应细胞低比例时可特异住抑制与其带有相同H-2单型的刺激细胞参与组合的增殖应谷和杀伤效应的诱导;在与反应细胞高比例时其抑制不具特异住,但对与其带有相同H-2单型的刺激细胞参与组合的抑制较强,提示LABMC在低比例时具Veto活性,在高比例时同时具Veto和自然抑制(NS)活性。LABMc于培养第二天加入单向MLC则不显示抑制效应,提示LABMC对MLC的抑制具有时相效应,流式细胞术(FCM)显示LABMC中lyt-2(+)细胞成分明显增加. MLR-CML model was established successfully by using oneway MLC technique in a va-riety of experimental combinations of inbred strains with different H-2 haplotypes to observe theveto activity of LABMC. Results showed that:(1)LABMC added to MLC on day 0 specificallyinhibited the proliferation response and cytotoxic effect in MLC in which stimulator cells havethe same H-2 haplotypes with LABMC at the LABMC/responder cell ratio of 1/10, but non-specifically suppressed the proliferation response and cytotoxic effect in MLC in vvhich stimulatorcells have the same or different H-2 haplotypes with LABMC at the higher LABMC/respondercell ratios of 1/3,1/1 and 3/1.Suggesting that LABMC have veto activity at the lower ratios,but have veto and natural suppressor(NS)activity at higher ratios.(2)LABMC added to MLCon day 2 had no suppressive activity. suggesting that the suppression of LABMC on MLC is re-lated to phase, (3)Phenotype analysis by flow cytotnetry showed that lyt-2(+)cell increased inLABMC。
