辛伐他汀与罗格列酮对动脉粥样硬化大鼠CD40和CD40L表达的影响 被引量:6

摘要 目的 观察辛伐他汀与罗格列酮对动脉粥样硬化大鼠CD40 和CD40L表达的影响。方法 通过高脂饮食和采用维生素D负荷法复制早期动脉粥样硬化大鼠模型,分别予辛伐他汀、罗格列酮(剂量分别为10 mg·kg-1·d-1、2.5 mg·kg-1·d-1,n=10)干预;采用酶法测定大鼠血脂、苏木精伊红(HE)染色观察大鼠胸主动脉病理变化、半定量逆转录聚合酶链反应检测大鼠腹腔巨噬细胞CD40 和CD40L mRNA的表达。结果 动脉粥样硬化组大鼠主动脉出现形成纤维增生性动脉硬化斑块,经辛伐他汀和罗格列酮干预后病变程度减轻;与动脉粥样硬化组相比,辛伐他汀或罗格列酮干预后的大鼠血总胆固醇均显著下降(P<0.01),低密度脂蛋白胆固醇逆转录聚合酶链反应及三酰甘油亦明显降低(P<0.05);CD40 和CD40L mRNA表达分别由0.96±0.24降至0.76±0.15和0.89±0.22、由0.73±0.18降至0.62±0.17和0.54±0.12(P<0.05)。结论 辛伐他汀与罗格列酮均能降低血脂,并通过降低巨噬细胞CD40和CD40L表达发挥抗动脉粥样硬化作用。 Objective To observe the effects of simvastatin and rosiglitazone on the expressions of CD40 and CD40L in perilioneal macrophages of atherosclerotic rats. Methods Early atherosclerotic rat model was constructed experimentally by high-fat diet and vitamin D loading, and simvastatin or rosiglitazone was taken for therapeutic intervention (10 mg/kg or 2.5 mg/kg body weight everyday for 2 weeks,respectively, n=10). The pathological changes of thoracic aorta by hematoxylin and eosin staining, the serum lipids of rats through enzymatic approach and the expression of CD40 and CD40 ligand mRNA with semi-quantitive RT-PCR in perilioneal macrophages of rats were undertaken. Results Fibrous hyperplastic plaques were shown on the aorta of arteriosclerotic rat, which were ameliorated following the administration of simvastatin or rosiglitazone. The serum levels of total cholesterol, low-density lipoprotein-cholesterol, and triglyceride decreased significantly in the treated group comparing with those in arteriosclerotic one without drug administrution P< 0.01 and P< 0.05 respectively. RT-PCR showed that CD40 mRNA decreased from 0.96±0.24 in the arteriosclerotic rats to 0.76±0.1 or 0.89±0.22 in simvastatin or rosiglitazone group respectively, and CD40 ligand from 0.73±0.18 to 0.62±0.17 or 0.54±0.12(P< 0.05). Conclusion Simvastatin and rosiglitazone can decrease serum level of lipids and exert anti-atherosclerotic effect by reducing the expression of CD40 and CD40L.
