摘要 目的 观察病毒性心肌炎(VMC)的小鼠模型中肿瘤坏死因子(TNF) -α和Fas mRNA在心肌中表达的动态变化,探讨它们在VMC中的可能作用及其机制。方法 腹腔接种柯萨奇病毒B组3 型(CVB3 )建立VMC小鼠模型。在接种病毒后第7、14 和28 天,采用逆转录聚合酶链反应(RT- PCR)检测小鼠心肌中TNF -α和Fas mRNA表达水平,并观察心肌形态学改变的差别。结果 与正常对照组相比,VMC小鼠TNF -α和FasmRNA表达明显增加。TNF- αmRNA表达高峰在接种后第7天,14 d后仍有大量表达,28 d后表达明显减少;Fas mRNA在接种第7天后明显增加, 14 d 后达到高峰, 28 d 后仍有一定量表达。结论 VMC小鼠心肌中TNF -α和Fas mRNA表达明显增加。TNF- α参与炎症反应和心肌损伤,Fas是引起心肌细胞坏死的重要蛋白。 Objective To study the changes of tumor necrosis factor-α and Fas mRNA in murine viral myocarditis (VMC), and to appraise their roles and implications in VMC. Methods Viral myocarditis model was constructed by intraperitoneal inoculation with CVB 3 solution. The expression levels of myocardial TNF-α and Fas mRNA in VMC mice were studied by RT-PCR, and myocardial histopathologic changes were observed. Results Compared with the normal control group, the expression of TNF-α and Fas mRNA increased remarkably. The peak expression of TNF-α mRNA reached at day7 after inoculation, and high level was maintained even after day14, but decreased significantly on day28. The expression of Fas mRNA increased on the 7th day after the inoculation, and reached the peak after the 14th day and still showed expression on the 28th day. Conclusions The expression of myocardial TNF-αand Fas mRNA increase obviously while TNF-α plays a role in the inflammatory reaction and myocardial damage, Fas takes part in myocardial necrosis of VMC.
