摘要
目的:探讨低密度脂蛋白免疫复合物(L DL- IC)对单核细胞衍生巨噬细胞中丙二醛(MDA)产生的影响。方法:采用比色法测定培养上清MDA含量。观察不同浓度L DL - IC和不同作用时间巨噬细胞MDA产生的变化,并设立终浓度均为15 0 μg/ ml的L DL- IC、L DL、Ig G- IC及阴性对照组与细胞孵育36 h,比较各组培养上清MDA水平的差异。结果:MDA水平随着L DL - IC的作用浓度与孵育时间增加而升高。与阴性组相比,L DL - IC、L DL及Ig G- IC均能明显提高上清MDA含量,而L DL- IC的作用最明显(P<0 .0 1)。结论:L DL- IC可以显著增加巨噬细胞MDA的产生,提示L DL - IC能增加活性氧物质而在动脉粥样硬化中起重要作用。
Objective:To study the effect of LDL-containing immune complexes(LDL-IC) on the malondialdehyde production in monocyte derived macrophages.Methods:To investigate whether there was any dose or time dependent effect on the MDA production,PMA-treated THP-1 cells were incubated with different concentrations of LDL-IC(0、50、100、150、200、250μg/ml)for 36h or treated with 150μg/ml LDL-IC for different hours(0、3、6、12、24、36h)。Moreover,the luminescence level of MDA of the four matched groups of LDL-IC,native LDL,immune complexes of IgG(IgG-IC) and negative control was measured respectively.Results:The MDA levels were correlated with both LDL-IC concentration and incubating time.The MDA levels of the macrophages incubated with LDL-IC,native LDL,IgG-IG were all elevated markedly compared to negative control(P<0.01),but the increased level of the group incubated with LDL-IC was the highest(P<0.01).Conclusions:LDL-IC have the greatest capacity to promote the production of MDA.LDL-IC may play an important role in the development of atherosclerosis by reactive oxygen species release.
