光生物素标记Patoc 1株DNA探针鉴别双曲和问号钩端螺旋体的初探

摘要 本文以光生物素标记的双曲钩体Patoc1株全DNA作为探针,对不同血清型的问号钩体与双曲钩体在硝酸纤维素膜上和微量反应板中进行DNA-DNA杂交试验。结果发现标记探针只能识别双曲钩体DNA,而对问号钩体DNA无一发生杂交,表明该探针具有高度特异性和敏感性,可用于鉴别双曲钩体和致病性钩体。 In this study,the whole DNA of L.biflexa sv Patoc strain Patoc 1 was labelled with photohiotin. It was used as a probe to detect different serovars of L. interrogans,and L. biflexa sv Andamana Strain CH11 by the dot-hybridization method on a nintrocellulose membrane and the microtiter plate method.The results showed that only the DNA of L. biflexa(strain CH11 and strain Patoc 1) could be recongnized by this probe. No hybridization was found between the DNA probe of strain Patoc 1 and all the tested pathogenic leptospiral DNA of various serovars.This finding indicated that photobiotin-DNA probe of strain Patoc 1 could be used to differentiate L.biflexa from L. interrogans.
