摘要
目的研究胚胎停止生长患者绒毛组织中的细胞凋亡及细胞凋亡调控基因bcl-2和bax在凋亡的作用。方法收集妊娠12周以内经B超证实为胚胎停止生长患者的绒毛组织30例;对照组30例为正常早孕要求人工流产者。所有研究对象均无生殖器畸形和感染性疾病,无内、外科合并症及妊娠并发症。应用TUNEL技术检测绒毛组织中的凋亡细胞,用免疫组织化学SP法观察细胞凋亡调控基因bcl-2和bax在绒毛组织中的表达。结果正常早孕和胚胎停止生长患者绒毛组织中均可见到凋亡细胞,但胚胎停止生长患者的绒毛组织中凋亡细胞显著增多。与正常早孕对照组比较有显著性差异(P<0.05);胚胎停止生长患者绒毛组织中可见bax过表达,与正常早孕对照组比较差异有显著性(P<0.05);而bcl-2在胚胎停止生长患者绒毛组织中表达低于正常对照组,两者比较差异有显著性(P<0.05)。结论胚胎停止生长患者绒毛组织中细胞凋亡指数显著增加且与bax表达呈正相关,而与bcl-2的表达呈负相关。
Objective To detect the effect of cell apoptosis and regulator gene (bcl-2 and bax) in the villosity tissue of dysontogenesis. Methods 30 cases of dysontogenesis were included in the experiment group, they were proved by B-ultrasound; control group had 30 cases,demanding the artificial abortion in early pregnancy. All the cases had no genital abnormality, infection diseases, pregnancy complication, medical and surgical complication. TUNEL was used to detect the apoptosis of cell, immunohistochemistry was used to measure the expression of bcl-2 and bax. Results Apoptosis cells could be found in normal pregnancy villosity and dysontogenesis, but there were more in the experiment group than that in the control group, the difference was significant(P<0.05);bax over-expressed in the experiment group ,there was significant difference compared to the control group(P<0 05).But the expression of bcl-2 were lower in the experiment group than that in the control group, there were significant difference between them(P<0.05).Conclusion The index of apoptosis in dysontogenesis increased significantly, and positively correlate to bax expression, but negatively correlate to bcl-2.
