摘要 目的分离纯化枸杞子多肽(PFL),探讨其组分1(PFL-1)对HT29细胞的作用。方法应用SpherisorC18色谱柱和乙腈溶液组成的HPLC色谱系统,检测波长214nm,对枸杞子多肽(PFL)进行分离,并对此多肽的各组分进行分峰收集;对所收集的各组分进行纯度分析。用MTT比色法测定了不同剂量的PFL-1对HT29细胞的抑制率。结果由PFL可分得三种组分,分别命名为PFL-1、PFL-2、PFL-3;PFL-1可使HT29细胞增值减弱,并与浓度呈正相关。结论PFL由三种成分组成,PFL-1对HT29细胞有抑制作用。 Objective To isolate and purify the polypeptide from Fructus lycii (PFL), and investigate the activity of component 1 (PFL-1) to HT29 cells. Methods Isolation of PFL was performed with an HPLC system: column of Spherisor C 18, mobile phase of acetonitrile and detected at 214nm. Every components of PFL were collected according to their peaks and analyzed for purity. The inhibitory rate of different dose of PFL-1 on HT29 cells were determined by MTT colorimertry method. Results Three components were obtained and named PFL-1, PFL-2 and PFL-3 respectively. When exposed to PFL-1, the proliferative ability of HT29 cell was dosage dependent. Conclusion PFL consists of three components, of which PFL-1 has inhibitory activity against HT29 cells.
