摘要
本文对IL-2基因工程菌的发酵及其包含体制备工艺进行了研究.结果表明在发酵过程中pH的稳定(7.0)、糖的补充(3%)、O2的及时供给(<10PSIG持续泵入)和发酵密度的控制(OD550=1.9),对提高发酵产量至关重要(12g/L).在包含体制备及溶解工艺中,以4mol/L脲去除脂质及杂蛋白,1%SDS溶解包含体,可获最大IL2活性单位(2.35×107u/ml,即相当于5L发酵可获3.53×109u).
The study on the technology of fermentation with 5 liter BIOFLO IIc Fermentor (New Brunswick Scientific Co, Inc) and preparation of inclusion body were made. The results showed that the factors including pH stability (7. 0), glucose addition (3%), DO (dissolve oxygen) supplementary (<10 PS1G continuesly added) and controlled cell density (OD500=1.9) played an important role in fermentation. In order to obtain IL-2 of highest activity, 4 mol/L urea was added to remove lipid and cell membranes and 1% SDS was added to dissolve inclusion body. By using this procedure, cell wet weight reached 12 g/L and IL-2 activity increased to 2. 35 107 u/ml (eg. 150 ml×2. 35×10 7 u/ml/5 liter fermentor).
