抗小儿白血病生物导弹的体外实验

摘要 建立了两株分泌抗人早幼粒细胞白血病细胞单克隆抗体细胞株,其单克隆抗体效价稳定,高达1:104。以此单抗作载体,高三尖杉酯碱作弹头,用化学方法偶联制成抗小儿白血病生物导弹,用UV谱和CD谱证实偶联成功。体外试验对靶细胞杀伤率达91.70%~97.98%,高于高三尖杉酯碱对靶细胞杀伤率,对人体各种正常细胞无损伤。 2 cell strains secreting McAbs to human promyelocytic leukemia were established and the secreted McAbs showed high (1: 104) and stable titers. Missile drugs for inffantile leukemia were prepared by chemical coupling, using the McAbs and homoharingtomine as carrier and warhead, respectively. UV and CD spectroscopy proved that the coupling was successful. Test in vitro showed that the lethal rate (91. 70%~97. 98%) of missile drug to target cellswas higher than that using homoharringtonine alone, and no impairment to normal cells was found.
