斑蝥素抑制NF-κB(P65)及Smad3在高转移卵巢癌细胞株HO-8910PM中的表达 被引量:14

摘要 目的:研究斑蝥素对H O- 8910 PM细胞内N F- κB(P6 5 )、Smad3蛋白及NF- κB(P6 5 ) m RN A表达的影响,探讨其抗肿瘤侵袭转移的作用机制。方法:用Western blot分析N F- κB(P6 5 )及Smad3蛋白的表达;RT- PCR分析NF- κB(P6 5 ) m RNA的表达。结果:2 5~10 0μmol/ L的斑蝥素作用于H O- 8910 PM细胞2 4 h后,明显下调N F-κB(P6 5 )及Smad3蛋白表达和NF-κB(P6 5 ) m RN A表达(P<0 .0 1) ,并呈一定的剂量效应。结论:斑蝥素抗肿瘤侵袭、转移与抑制NF- κB(P6 5 )、Smad3蛋白和NF- κB(P6 5 ) m RNA的表达有关。 Objective: To observe the effects of cantharidin on the expression of NF-κB(P65) and Smad3 genes in the ovarian carcinoma cell line HO-8910PM with highly metastatic potential in order to elucidate the mechanisms in which cantharidin inhibits the invasion and metastasis of tumor. Methods: The Western blot analysis was used to detect the protein levels of NF-κB(P65) and Smad3, and the RT-PCR assay was used to assess the mRNA level of NF-κB(P65). Results: The expression levels of NF-κB(P65) and Smad3 proteins and NF-kB(P65) mRNA were induced to decrease obviously in HO-8910PM cells by the treatment with 5~20 μmol/L of cantharidin for 24 hours. Conclusion: The cantharidin's activity against tumor invasion and metastasis are involved in the reduction of expression level of NF-κB(P65) and Smad3 protein and NF-κB(P65) mRNA .
