原花青素对脂多糖诱导的RAW264.7细胞NF-κB DNA结合活性的抑制作用 被引量:4

摘要 目的:观察原花青素对脂多糖(L PS)诱导的RAW2 6 4 .7细胞核转录因子- κB(nuclear factor Kappa B,NF- κB)结合活性的影响。方法:L PS诱导RAW2 6 4 .7细胞,加入不同质量体积浓度原花青素孵育1h,提取核蛋白,电泳迁移率变动分析检测NF-κB与DNA的结合活性。结果:L PS诱导小鼠巨噬细胞肿瘤细胞株RAW2 6 4 .7后,其NF-κB的核结合活性明显增高。0 .8、4和2 0 mg/L的原花青素可明显降低L PS诱导下RAW2 6 4 .7细胞的NF-κB活化。结论:原花青素对NF-κB活化的抑制作用可能是其抗炎的作用机理之一。 Objective: To observe the effect of proanthocyanidins on the LPS-induced DNA-binding activity of nuclear factor of kappa B(NF-κB) in RAW 264.7 cells. Methods: After different concentrations of proanthocyanidins were added into the LPS-pretreated RAW264.7 cells in cell culture followed by the incubation for 1 hour, the nuclear protein was isolated from the cells and the DNA-binding activity of NF-κB from the isolated protein was measured by the electrophoretic mobility shift assay (EMSA). Results: The DNA-binding activity of NF-κB was increased significantly after the RAW 264.7 cells were stimulated with 1 mg/L of LPS. The concentrations of proanthocyanidins at 0.8 mg/L, 4 mg/L and 20 mg/L reduced the DNA-binding activity of NF-κB in the LPS-pretreated RAW264.7 cells. Conclusion: The inhibitory effects of proanthocyanidins on the DNA-binding activity of NF-κB might be one of its functions in anti-inflammation.
