剪切力对脐静脉内皮细胞凋亡的影响 被引量:3

摘要 内皮细胞的炎症反应和凋亡是动脉粥样硬化过程中的关键环节 ,因此通过建立HUVEC(人脐静脉内皮细胞 )体外培养模型 ,以LPS(脂多糖 )刺激 ,观察其损伤效应。并在此基础上施加两种大小剪切力 ,以探讨流体动力学对血管内皮细胞凋亡的影响。同时 ,观察此过程中IL 6及IL 8的变化。结果显示 ,LPS加入细胞培养中可明显诱导HUVEC的凋亡 ,并随着作用时间的延长 ,细胞凋亡率也相应增加。在此基础上 ,将两种大小剪切力与LPS共同作用于细胞 ,发现剪切力 (15dyn/cm2 )可以抑制LPS诱导凋亡 ,而剪切力 (4dyn/cm2 )效果则不明显。同时 ,剪切力与LPS这两种均能单独诱导IL 6及IL 8分泌的刺激因素并不能协同作用 ,剪切力会抑制LPS诱导的IL 6分泌。且剪切力 (15dyn/cm2 )比剪切力 (4dyn/cm2 )时的抑制作用要强。 No abstract available
