摘要
目的:建立测定石蒜中加兰他敏含量的HPLC方法。方法:采用ODS色谱柱(46mm×150mm,5μm),以磷酸盐缓冲液(pH3~4)甲醇(93∶7)为流动相,流速10mL·min-1,二极管阵列检测器于289nm测定。结果:加兰他敏在3~30μg·mL-1线性关系良好(r=0.9997),最低检测限为0.30ng,测定加兰他敏的平均回收率为99.5%,RSD0.5%。结论:方法操作简便,结果可靠,重复性好,专属性强,将为石蒜的质量控制提供科学依据。
Objective: To establish a method for determination of galanthamine in Lycoris radiata . Method: HPLC separation was carried on a column of ODS (4.6 mm×150 mm,5 μm),with the mobile phase of phosphate buffer (pH=3-4)-methanol (93∶7) at the flow rate of 1.0 mL·min -1 ,and the detection wavelength was set at 289 nm. Result: Galanthamine revealed linearity within the range of 3-30 μg·mL -1 ( r =0.999 7), the detection limit was 0.3 ng. The average recovery was 99.5%(RSD=0.5%). Conclusion: The method is easy to operate and the results of the determination are accurate, it can be used to evaluate the quality of L. radiata .
