摘要
用纳米金溶胶与聚乙烯醇缩丁醛(PVB)构成复合固酶基质,采用溶胶凝胶法固定葡萄糖氧化酶(GOx)于铂金电极表面,并在葡萄糖溶液中加入天青Ⅰ作为电子媒介体,制成了新型葡萄糖生物传感器。实验证明,葡萄糖氧化酶吸附在纳米金颗粒表面上稳定且保持其生物活性;而电子媒介体的存在,显著提高了传感器的响应灵敏度。该传感器对葡萄糖响应的线性范围为2.5×10-5~7.5×10-3mol/L;检出限为8.5×10-6mol/L(S/N=3)。该生物传感器用于人体血清中的葡萄糖测定,结果令人满意。
This paper describes the fabrication and evaluation of a new approach to design and use of enzyme-based sensor systems. Glucose oxidase (GOx) adsorbed to colloidal gold sots combined with polyvinyl butyral (PVB) was immobilized onto the surface of a platinum electrode to make a glucose biosensor. The GOx immobilized on colloidal gold was stable and retained its functional activity. With the aid of azure as a mediator, the biosensor was substantially improved sensitivity and detection limit toward the glucose. Moreover, the relatively lower operation potential can effectively eliminate interference from common interferents. The oxidation current investigated with cyclic voltammetric peak current was linear with glucose concentration in the range of 2.5 x 10(-5) mol/L to 7.5 x 10(-3) mol/L with a detection limit of 8.5 X 10(-6) mol/L.
