GAD及GADmRNA在人胃癌组织中的表达

GAD and GADmRNA expressions in human gastric cancer

摘要 目的探讨人胃癌谷氨酸脱羧酶(GAD)、GADmRNA(包括GAD65mRNA和GAD67mRNA)的表达。方法 随机选取30例胃癌手术标本,采用免疫组化法捡测GAD,RT-PCR法检测GADmRNA的表达,并探讨与胃癌部位、浸润深度、分化程度、分期及有无淋巴结转移之间的关系。结果与正常胃黏膜组织比较,胃癌组织中GAD表达减弱(p<0.05),但与胃癌的临床病理特征无明显相关,人胃癌组织及正常胃黏膜组织只有GAD67mRNA的表达,而无GAD65mRNA的表达。结论GAD表达异常可能与胃癌的发生有密切关系;人胃癌组织及正常胃黏膜组织只有GAD67mRNA的表达。 Objective To explore the role of glutamate decarboxylase (GAD) in the pathogenesis of human gastric cancer. Methods Thirty patients with primary gastric cancers who were diagnosed and treated at our hospital were enrolled into the current study. Immunohistochemistry was carried out to check the ex-pression of GAD protein. Reverse transcription polymerase chain reaction ( RT-PCR) was performed to ex-amine the expression of GADmRNA. Then investigated the relationships among the sites of gastric cancer, depth of infiltration, and degree of differentiation, staging and lymphatic metastasis. Results Immunohisto-chemistry analysis showed that GAD expression decreased in gastric cancer tissues compared with that of ad-jacent normal tissue(P <0. 05). In human gastric cancer tissue, the wick positive expression of GAD was not prominently correlated with the clinical-pathological character of the cancer (P < 0. 05). RT-PCR analy-sis showed that the gastric mucosa was GAD65-negative and GAD67- positive. Conclusion GAD67mRNA predominates in human gastric mucosa and the abnormalities of GAD expression might contribute to the path-ogenesis of human gastric cancer.
