摘要
目的探讨建立一种简便、快捷检测人体纤维蛋白溶解(纤溶)功能的新方法。方法利用UV2000型分光光度计联接微机,自动扫描观测并记录血液凝固及溶解的光密度动态图形改变过程,并以此了解血浆抗、促纤溶的能力。结果初步建立了正常人纤溶动力学分析参数,确定了20U/ml尿激酶为最适试验浓度,检测22例正常人,27例急性髓细胞白血病患者纤溶动力学及纤溶酶原含量,显示患病组较正常组差异有显著意义(P<0.05,P<0.01),重复性试验显示该方法的变异系数为(5.42±1.51)%,并可同步测定血浆纤维蛋白原浓度。结论据结果可认为该方法稳定可靠,可为临床提供动态、直观、多参数的实验资料,在评价血浆抗、促纤溶能力、治疗监测、药物选择、浓度调节等方面均有价值。
Objective To explore a new,convenient and rapid method for kinetic measrement of human fibrinolysis. Methods A UV2000 spectrophotometer was used to connect with a computer,automatically scanning and recording the alteration of optical density in the process of blood coagulation and lysis. Results We established the parameter of human fibrinolysis kinetics.Results showed that urokinase at 20U/ml was the optimal concentration used.Fibrinolysis kinetics and plasma plasminogen concentration of 27 normal persons and 27 patients with acute myeloblastic leukemia were detected, a significant difference was observed between the two groups.Repetition test showed that the coefficience of variation was 5.24±1.51%. This method could also be used to detect the concentration of plasma fibrinogen.Conclusions The method was stable and could provide dynamic,direct experimental data and multiparemeters for clinicians. It is also valuable in evaluating the anti-or pro-fibrinolytic capacity of patient plasmas and in monitoring of therapy,drug selection and adjustment of drug concentrations.
