摘要
目的 研究丙酯对缺氧性血管内皮细胞凋亡的影响。 方法 用氰化钠(NaCN)合并低糖DMEM培养液构建培养的人脐静脉血管内皮细胞株(ECV30 4 )缺氧模型;四唑盐法、丫啶橙/溴乙啶荧光染色、末端脱氧核糖核酸转移酶介导的dUTP缺口末端标记等观察细胞受损和凋亡情况。 结果 NaCN合并低糖培养液可引起血管内皮细胞凋亡。4 μg/mL丙酯可显著减少由于NaCN引起的缺氧性内皮细胞凋亡(P <0 .0 5 )。 结论 丙酯对缺氧条件下的ECV30 4细胞具有保护作用。
Objective To study the effect of propylgallate on vascular endothelial cells apoptosis induced by chemical hypoxia-ischemia in vitro. Methods The chemical hypoxia-ischemia model was performed by treating cultured vascular endothelial cells(ECV304) with NaCN in low-glucose medium. Cell viability was determined by MTT. Cell apoptosis was defined by AO/EB staining and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling(TUNEL). Results Chemical hypoxia-ischemia was shown to initiate vascular endothelial cells apoptosis. Propylgallate(2~4 μg/mL) inhibit endothelial cell apoptosis effectively. Conclusion Propylgallate protect endothelial cells from apoptotic induced by chemical hypoxia-ischemia.
