摘要
目的:研究Isogoniotriol衍生物对肿瘤细胞DNA的损伤作用。方法:用Isogoniotriol衍生物处理体外培养白血病K562细胞,MTT法测定IC50,应用单细胞凝胶电泳(SCGE)检测细胞核DNA的损伤。结果:Isogoniotriol衍生物处理K562细胞IC50为4×10-5mol/L,不同浓度的Isogoniotriol衍生物处理细胞后可见彗星状拖尾。结论:Isogoniotriol衍生物可造成肿瘤细胞DNA损伤,抑制肿瘤细胞生长。
Objective: To investigate the damage of tumor cell DNA caused by Isogoniotriol derivatives. Methods: K562 cells were cultured in the experiment. The damage of tumor cell DNA was studied by single-cell gel electrophoresis. IC50 was measured by MTT. Results: K562 cells were treated with Isogoniotriol derivatives in different concentrations and the IC50 values were 4 × 10-5 mol/L. Single-cell gel electrophoresis showed comet-like tails. Conclusion: Isogoniotriol derivatives can damage DNA and inhibit proliferation of tumor cells.
