摘要
目的:探讨非小细胞肺癌(NSCLC)细胞凋亡与相关基因bcl-2、bax表达产物的关系及其临床意义。方法:应用流式细胞术和间接免疫荧光标记法进行检测。结果:凋亡率,Bcl-2、Bax蛋白标记率在NSCLC组与对照组中的差别有统计学意义(P<0.05)。NSCLCI期凋亡率明显高于II、III期组(P<0.01)。NSCLCI期组Bcl-2蛋白标记率明显低于II、III期组(P<0.01)。NSCLC中Bcl-2蛋白标记率与凋亡率呈负相关(r=-0.667,P<0.01)。Bax标记率与凋亡率无直线相关性(r=0.230,P>0.05)。Bcl-2/Bax>1组凋亡率明显低于Bcl-2/Bax≤1组(P<0.05)。结论:随着NSCLC临床分期的进展凋亡率下降,Bcl-2通过抑制凋亡影响NSCLC的进展。NSCLC细胞内存在一种对死亡讯号的应答模式:bcl-2/bax。
Objective: The aim of this study was to observe the relationship between apoptosis and related gene expression of bcl-2, bax and their clinical significance in non-small-cell lung cancer (NSCLC). Methods: The flow cytometry technique and indirect immunofluorescence staining were used in this study. Results: The apoptosis rate as well as the positive rate of Bcl-2 and Bax in the NSCLC group was significantly different (P < 0.05) from those in the control group. The apoptosis rate in stage I NSCLC was significantly higher than that in stage II-III NSCLC (P < 0.01). The percentage of Bcl-2 positive cells in stage I NSCLC was significantly lower than that in stage II-III NSCLC (P < 0.01). The percentage of Bcl-2 positive cells was inversely correlated with apoptosis rate (r = -0.667, P < 0.01). There was no correlation between the percentage of Bax positive cells and apoptosis rate (r = 0.230, P > 0.05). The apoptosis rate in the groups of Bcl-2/Bax > 1 was significantly lower than that in the group of Bcl-2/Bax ≤ 1. Conclusion: With the development of stage in NSCLC, the apoptosis rate decreases, which implies apoptosis rate may be correlated with the progression of NSCLC. Bcl-2 protein enhances growth rate of NSCLC due to blockage of cell death. The model with the ratio of bcl-2 to bax can determine survival or death following an apoptotic stimulus.
