摘要
目的:研究ERK在TGF - β1刺激肝癌细胞表达血管内皮生长因子中的作用。方法:体外培养肝癌细胞系HepG2 ,施加外源性TGF - β1,并使用细胞外信号调节激酶ERK1/2的特异性阻断剂。免疫组化以及RT -PCR检测VEGF蛋白和mRNA的表达。结果:与空白对照组比较,施加外源性TGF - β1能够使肝癌细胞VEGF的蛋白及mRNA的表达显著增多,但是同时使用阻断剂组的VEGFmRNA表达与TGF组相比没有明显差异。结论:TGF - β1刺激肝癌细胞HepG2表达VEGF可能通过ERK/MAPK以外的信号通路实现。
Objective:To observe the signal transduction pathway of vascular Endothelial Growth Factor(VEGF) transcription in hepatoma cells affected by TGF-β_1.Methods:HepG2 cells were cultured invitro,and treated with TGF-β_1 and the inhibitors of ERK-specific mitogen activated protein kinase(MAPK/ERK1/2).The changes of VEGF protein and mRNA were detected by immunohistochemistry and reverse transcriptase-polymerase chain reaction (RT-PCR). Resultes:Immunohistochemistry and RT-PCR showed that the expression of VEGF protein and mRNA were significantly higher in the group cultured with TGF-β_1 than that of control groups.But there was no remarkable discrepancy in PD98059+TGF-β_1group compared with TGF-β_1group.Conclusion:Expression of VEGF stimulated by TGF-β_1 in HepG2 cells may be achieved through another signal pathway instead ERK/MAPK pathway.
