单抗Mab03.2C1C2影响白念珠菌芽管形成及粘附作用的实验研究 被引量:3

摘要 目的 观察抗白念珠菌芽管单抗Mab0 3 .2C1C2在体外对白念珠菌感染的保护性作用。方法 将不同浓度的Mab0 3 .2C1C2分别与白念珠菌孢子悬液、白念珠菌孢子和人口腔粘膜上皮细胞混合液、白念珠菌孢子和胎儿脐静脉内皮细胞混合液共同孵育,观察Mab0 3 .2C1C2对白念珠菌芽管生成的抑制作用及其对白念珠菌对上皮细胞、内皮细胞粘附的抑制作用。结果 Mab0 3 .2C1C2能显著降低白念珠菌芽管生成率及在芽管形成条件下对上皮细胞、内皮细胞的粘附,其抑制作用与该单抗的浓度成正比。结论 Mab0 3 .2C1C2在体外能抑制白念珠菌芽管的形成,通过抑制白念珠菌芽管的形成而抑制白念珠菌对上皮细胞、内皮细胞的粘附,降低白念珠菌的侵袭力。 Objective To investigate protective effects of monoclonal antibody Mab03.2C1C2 specific against C. albicans germ tube. Methods The different concentrations of Mab03.2C1C2 was incubated with C. albicans spores, the mixed suspenion of C. albicans spores and human buccal epithelial cells, the mixed suspenion of C. albicans spores and endothelial cells of fetal umbilical vein. Results In the protective Mab screening experiment, it was found that Mab03.2C1C2 could inhibit C. albicans germ tube formation and adhesion of C. albicans to epithelial and endothelial cells. Conclusion Mab03.2C1C2 could inhibit the adhesion of C. albicans to the epithelial and endothelial cells and decrease the invasion of C. albicans due to its inhibition effects on the formation of C. albicans germ tube.
