人胚胎胸腺基质细胞的体外分离培养与鉴定

摘要 目的:体外分离培养胎儿胸腺基质细胞,即上皮网状细胞,为研究T淋巴细胞的分化增殖及相关药物筛选奠定基础。方法:从水囊引产的胎儿胸腺中分离组织细胞,经筛选培养获得胸腺上皮网状细胞。结果:细胞染色体正常,为46条染色体,包括1对性染色体。倒置显微镜下观察,细胞呈扁平状,多边形,胞体较大,核圆形或椭圆形,核仁明显,胞质内含明显的颗粒和小泡。透射电镜显示细胞间有桥粒相连,并可见张力丝。用免疫组化法检测上皮细胞特异性表达标志(角蛋白)为阳性。结论:本法获得细胞为胸腺网状上皮细胞,为进一步工作打下基础。 Objective: To acquire thymus stroma cells from fetal thymus as the foundation for the study on differentiation and proliferation of T lymphocyte and screening of medicines. Methods: The thymus tissue cells were isolated from embryo thymus. After screening and culture, the reticular epithelial cells were obtained. Results: The cells were flat,polygonal and large. They had round or oval cell nuclei, in which the nucleoli were evident, and vesicles and granules in cytoplasm. Electron microscopic observation indicated that the cells contained tonofibrils and desmosomes. Immunohistochemical examination showed that keratin was expressed. The cell chromosome was normal. Conclusion: The reticular epithelial cells from fetal thymus were obtained.
