血管内皮细胞相关生长因子在高血压不同阶段心肌细胞和血管内皮细胞中的分布特征 被引量:3

摘要 目的:观察血管内皮细胞生长因子、碱性成纤维生长因子在高血压不同阶段心肌细胞和血管内皮细胞的分布变化。方法:实验于2001-10/2002-06在承德医学院附属医院中心实验室完成。选用遗传性盐敏感型高血压大鼠(DahlS)和遗传性盐抵抗型高血压大鼠(DahlR),在5周龄投与8%食盐饲料,分别在10,11,12周时取材,制作心肌组织切片。应用免疫组织化学方法观察血管内皮细胞生长因子和碱性成纤维生长因子在心肌组织中的表达变化情况,计算单位面积心肌组织血管内皮细胞生长因子和碱性成纤维生长因子的阳性细胞数和血管内皮细胞的阳性表达。结果:血管内皮细胞生长因子和碱性成纤维生长因子免疫细胞在DahlS和DahlR组大鼠的心肌细胞及血管内皮细胞中均有表达,DahlS组血管内皮细胞生长因子和碱性成纤维生长因子阳性细胞数均较同时期DahlR组增多(P<0.01)。DahlS组自10周起至12周心肌细胞血管内皮细胞生长因子和碱性成纤维生长因子阳性细胞数逐渐增多(P<0.01)。DahlR组在高血压不同时期血管内皮细胞生长因子和碱性成纤维生长因子无显著性变化(P>0.05)。心肌组织的血管内皮细胞的表达强度,DahlS组血管内皮细胞生长因子和碱性成纤维生长因子均由(+)到(),且均可见组织内毛细血管增多。DahlR组无变化。结论:血管内皮细胞生长因子和? AIM:To observe the distribution of vascular endothelial growth factor(VEGF) and basic fibroblast growth factor(bFGF) in cardiocytes and vascular endothelial cells(VECs) at the different stages of hypertension. METHODS:The experiment was conducted in the central laboratory of the Affiliated Hospital of Chengde Medical College from October 2001 to June 2002.The genetic salt-sensitive hypertension rats(Dahl-S) and the genetic salt-resistive hypertension rats(Dahl-R) were chosen as the subjects.When they were 5 weeks old,they were fed with 8%salt content feedstuff.The materials were drawn respectively from rats at the age of 10,11 and 12 weeks to make sections of myocardial tissues.With the immunohistochemical technique,the changes of VEGF and bFGF expression in myocardial tissues were observed.Positive cells of VEGF and bFGF in the myocardial tissues were counted within each area unit and the positive expression of VECs was calculated. RESULTS:VEGF and bFGF immunocytes expressed in cardiocytes and VECs in both Dahl-S rat group and Dahl-R rat group.The amount of VEGF and bFGF positive cells was significantly greater in Dahl-S rat group than in Dahl-R rat group at the same stage(P< 0.01).The number of VEGF and bFGF positive cells was gradually increased in Dahl-S rat group from 10 to 12 weeks(P< 0.01).VEGF and bFGF had no significant changes in Dahl-R rat group at the different stages of hypertension(P >0.05).In Dahl-S rat group,the intensity of expression of VECs in VEGF and bFGF increased from(+) to(),and the capillary density in tissues was found increased.It was unchanged in Dahl-R rat group at any stage. CONCLUSION:There is expression of VEGF and bFGF in both cardiocytes and VECs and the trend is going upwards as the hypertension goes on.
