衰老大鼠卵巢Bax及血管内皮生长因子表达与滋阴理气中药的干预效应 被引量:2

Effects of Chinese herb of nourishing yin and regulating qi on the expressions of Bax and vascular endothelial growth factor in ovary of D-galactose induced aging rats

摘要 目的:研究衰老大鼠卵巢中Bax和血管内皮生长因子的表达及滋阴理气中药对其表达的影响,探讨滋阴理气中药延缓衰老的分子机制。方法:实验时间:2004-02/09。实验地点:哈尔滨医科大学生理教研室实验室。将24只2.5个月龄雌性SD大鼠随机分为3组:对照组、模型组和中药组,每组8只。通过腹腔注射D-半乳糖建立衰老大鼠模型,应用反转录聚合酶链反应和免疫组化SP法检测各组卵巢组织中BaxmRNA及血管内皮生长因子蛋白的表达。结果:与对照组相比,模型组卵巢组织BaxmRNA表达增强,血管内皮生长因子蛋白表达阳性目标PU值(7.75±2.31)较对照组PU值(4.86±1.52)增大(P<0.01);与模型组相比,中药组可下调BaxmR-NA的表达,血管内皮生长因子蛋白表达阳性目标PU值(5.93±1.78)较模型组(7.75±2.31)减小(P<0.05)。结论:D-半乳糖造模可引起卵巢BaxmRNA和血管内皮生长因子蛋白表达上调,滋阴理气中药可下调BaxmRNA和血管内皮生长因子蛋白的表达,这可能是滋阴理气中药延缓卵巢老化的机制之一。 AIM: To investigate the expressions of Bax and vascular endothelial growth factor (VEGF) in ovary of aging rats and the effect of Chinese herb of nourishing yin and regulating qi, and probe into the molecular mechanism of the Chinese herb in delaying aging. METHODS: The experiment was carried out in the Department of Physiology, Harbin Medical University from February to September 2004. Twenty-four 2.5-month-old female SD rats were randomly divided into control group (n=8), model group (n=8) and Chinese herb group (n=8). Aging rat models were established by intraperitoneal injection of D-galactose, and the expressions of Bax mRNA and VEGF protein in ovary were examined with reverse transcriptase polymerase chain reaction (RT-PCR) and immunohistochemical SP technique. RESULTS: Compared with the control group, the expression of Bax mRNA in ovary was enhanced in the model group, and positive PU value of VEGF protein was higher (7.75±2.31 vs 4.86±1.52) (P< 0.01). Compared with model group, the expression of Bax mRNA in ovary was down-regulated, and the positive PU value of VEGF protein was reduced (5.93±1.78 vs 7.75±2.31)(P< 0.05). CONCLUSION: The expressions of Bax mRNA and VEGF protein in ovary are up-regulated in D-galactose induced aging rats. The Chinese herb of nourishing yin and regulating qi can down-regulate the expressions of Bax mRNA and VEGF protein, which may be one of the mechanisms of delaying ovary aging by the Chinese herb.
