摘要
以橄榄叶片为材料,进行橄榄基因组DNA提取及RAPD扩增条件优化。试验表明,采用改良的SDS法,提取的DNA质量较高,适宜于RAPD分析。在25μL反应体积中,RAPD反应的优化体系为:18.5μLddH2O,0.5μLdNTP,2.5μlBuffer,1μL引物,0.5μLDNA模板,2μLTaqDNA聚合酶。
The method of genomic DNA extraction and the optimization of RAPD analytic condi tions were studied in Canarium album Reausch.The result showed that the high -quality genomic DNA was obtained by the revised SDS method. The optimal PCR sys tem for RAPD analysis was as follows: 18.5μL dd H_2O, 0. 5μL dNTP, 2.5μL Buffer,1μL random primer,0.5μL tem plate,1u Taq polymerase in 25μL reaction system.
