摘要
应用抗玉米赤霉烯酮单克隆抗体(ZEN-McAb)建立了竞争间接ELISA,用以检测玉米、小麦、饲料中的玉米赤霉烯酮(ZEN),最低检出量为0.05ng/mL检测范围为0.05~500ng/mL。检测掺合ZEN(25~500ng/g)的小麦、饲料、玉米样品,平均回收率,玉米为105.8%、小麦为90%、饲料为103.9%;平均批内和批间变异系数,玉米为3.8%、5.8%,小麦为12.7%、28%,饲料为15.2%、6.8%。对玉米、小麦、饲料自然样品检测,竞争间接ELISA的检出率高于薄层色谱(TLC),检测程序简易快速。
A competitive indirect ELISA with anti-ZEN McAb for
detecting themycotoxin zearalenone in corn, wheat and feedstuffs was developed. The
minimum de-tectable amount was 0.05 ng/mL and the limits ranged from 0.05 ng to 500 ng
permillilitre。 In the detection of the samples of corn,wheat and feedstuffs mixed with
ZEN(2O-500 ng/g), the average rates of reclamation were 105.3%, 90%and 102.9%re-spectively.
The mean intra-batch and inter-batch coefficients of variation were 3.8%and 5.8%in
contaminated corn, 12.7%and 2.8%in contaminated wheat and 5.2%and 6.8%in contaminated
feedstuffs, Using the developed competitive indirect ELISAto detect ZEN in natural samples of
corn, wheat and feedstuffs , the detective rates washigher than those by thin-layer
chromatography。 The assay procedure is simple, quickand easy to perform。
