红叶李组织培养快繁技术 被引量:7

摘要 通过培养红叶李的不同外植体,建立适宜的组织培养条件和快繁技术,为红叶李的组培繁殖提供技术参考。采用MS为基本培养基,添加不同浓度的激素,以茎尖、腋芽、茎段和叶片作为外植体进行培养。结果表明:茎尖和腋芽既能诱导芽的生长又能诱导愈伤组织,且诱导效果较好,而茎段仅能诱导产生愈伤组织,叶片的诱导效果不明显。BA 1.0 mg/L+2,4-D 0.5-2.0 mg/L组合的诱导效果最佳,单芽在BA 1.0 mg/L+IBA 0.5~2.0 mg/L培养基中继代培养都能诱导产生丛芽,诱导率达70%。 The stem apex, stem, axillary bud and leaf blade of Prunus cerasifera were used for explants to culture in the MS mediums added different concentration of 6-BA, NAA and 2,4-D for induction of buds and callus. The results showed that the combinations of 6-BA and 2,4-D had better induction effects than combinations of 6-BA and NAA. The stem apex and axillary buds can be inducted to produce new buds or callus and had fairly good effects, but stem and leaf can only be inducted to produce callus and had poor effects. In all experimental culture mediums, the combination of 6-BA1.0mg/L+2,4-D0. 5-2.0 had the best induction effects. There was low induced ratio of the adventitious buds from the callus, but the induction ratio from single-bud to arbustive buds in the mediums of MS + BA1. 0 + IBA0. 5 and MS + BA1. 0 + IBA1.0 were all above 70%.
