Analysis method of two-dimensional gel electrophoresis images

目的:建立一套双向凝胶电泳图像的分析方法,为进一步分析和鉴定差异蛋白奠定基础。方法:提取肺癌组织和癌旁组织中的可溶性总蛋白,进行双向凝胶电泳,电泳图片由专用扫描仪获取后,应用图像分析软件(PDQuest7.1)进行蛋白差异点的表达情况分析。结果:二维电泳图片经过背景消减、点检测、点匹配等一系列分析后,得出了差异点。结论:PDQuest7.1软件可以用来快速有效地对生物样品之间蛋白质的差异进行分析。

Aim: To establish a two-dimensional gel electrophoresis image analysis method for analyzing the differentially expressed proteins. Methods: The whole soluble proteins of tissue of lung cancer and the controls were extracted by one step and separated using 2-dimensional gel electrophoresis.Acquire the image and analyze the images using the software PDQuest 7.1. Results: After the background subtraction,spot detection and matching,the protein spots which expressed differentially were obtained. Conclusion: The results of dimensional gel electrophoresis can be ananlyzed quickly and conveniently using PDQuest 7.1 gel image analysis software.
