胚胎干细胞自我更新相关信号转导途径及信号分子 被引量:4

摘要 胚胎干细胞(embryonic stem cells,ES cells)是从囊胚期内细胞团(inner cell mass,ICM)分离得到的,在体外具有无限或较长期的自我更新能力,并能在特定的诱导条件下分化成各种组织特异性细胞.鉴于此种特性,ES细胞在临床上具有极其广阔的应用前景.但是要将ES细胞成功的应用于临床,必须解决的首要问题是胚胎干细胞是如何实现自我更新及定向分化的.某些小鼠的ES(mES)细胞可以在LIF等外源性细胞因子的作用下在体外维持自我更新.研究表明,LIF维持自我更新的能力是通过LIF-LIFR-JAK-STAT3途径实现的,其中STAT3在该途径中起关键作用.Oct-3/4也是维持ES细胞多能性的重要分子之一,它与其辅助分子Sox2,Rox-1共同参与调节胚胎的正常发育.Nanog是最近发现的一个同源盒转录因子,被认为在ES细胞的全能性维持中起关键作用.此外,BMP,Wnt,ERK途径也在这一过程中起重要作用.本文中我们对近年来发现的有助于胚胎干细胞自我更新的相关信号转导通路做了概括综述,主要有LIF-STAT3途径、Oct-3/4和Nanog信号分子.虽然它们之间是通过怎样的模式相互影响和协调起作用还不是十分清楚,但为我们以后的研究指明了方向.
