摘要
目的:探讨中国汉族人载脂蛋白E基因多态性与高密度脂蛋白(HDL)亚类组成变化的关系。方法:采用聚合酶链反应限制性片段长度多态性和双向电泳免疫印迹检测法,分析335例中国汉族人的apoE基因型、HDL亚类组成及相对含量。结果:apoE等位基因频率分别为:ε2(8.96%),ε3(81.94%),ε4(9.10%)。等位基因ε2携带者血清TG较ε3携带者升高、apoE和apoE/CⅢ较ε3和ε4携带者升高,而等位基因ε2携带者血清LDLC和HDL3b较ε3携带者下降,其差异有统计学意义(P<0.05)。等位基因ε2携带者血清HDL2a较等位基因ε3携带者有升高的趋势,但差异无统计学意义(P>0.05)。结论:apoE基因多态性可能与血清HDL部分亚类的含量变化相关。
Objective:To investigate human apolipoprotein(apo) E polymorphism and its relationship with serum lipids, apolipoprotein and serum HDL subpopulations. Method:apoE genotype was assayed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The subpopulations of serum HDL were determined by two-dimensional gel electrophoresis conjunction with immunodetection method. Result:apoE 3/3 genotypes and allele ε3 frequency were both the highest with allele frequency ε2(8.96%), ε3(81.94%) and ε4(9.10%) respectively. The genotype of apoE2 had higher serum TG,apoE levels and apoE/aopCⅢ ratio as compared with the genotype of apoE 3 or apoE 4 (P< 0.05), but lower LDL-C and HDL 3b levels compared with the genotype of apoE 3 (P< 0.05).In additional,we also found that in apoE 2 carriers HDL 2a have the tendency of increase compared with apoE 3 carriers,but no significant difference was observed. Conclusion:apoE gene polymoephism may related to the distribution of HDL subpopulations.
