摘要
目的:建立蛇毒抗瘤蛋白生物活性测定方法。方法:用正交实验确定实验条件,求细胞5 0 %死亡时的样品浓度,即IC50 值。结果:此法使细胞死亡率与样品的Log浓度具有良好的线性关系(r =0 .95 15 ,P <0 .0 1) ,重复性实验结果:板内RSD值为12 .3% ,板间RSD值为16 .6 % ;不同实验次数RSD值为17.8% ,灵敏度为36 μg·L- 1,并能反应出蛇毒抗瘤蛋白的抗肿瘤特异性。结论:此方法特异,简便且具有良好的重复性,可以作为蛇毒抗瘤蛋白生物活性的测定方法。
AIM: To develop a method for determination of the biological activity of atranticance. METHODS: The conditions of the experiment were determined by orthogonal test, then IC_ 50 which was the concentration of the sample when the 50% death of cells was tested. RESULTS: The mortality rate had a good correlativity with Log consistency of the sample, (r= 0.9515 , P< 0.01 ). And the coefficients of variation of reproducibility of intra-, inter and different test assays were 12.3 %, 16.6 % and 17.8 %, respectively. CONCLUSION: This method is superfine, simple and highly reproducible. It can be used for determining the biological activity of atranticance.
