摘要
目的对中国四川省汉族代表性人群的DYS19(DNAYchromosomemicrosatellite19)位点进行遗传多态性研究。方法基因组DNA的提取采用传统的酚-氯仿抽提法。DYS19位点的遗传多样性分析采用ABIPrismTM310遗传分析仪及扫描分析系统的微卫星基因组扫描分析。结果在四川省汉族群体中共发现拷贝数13~17五种等位基因型,其等位基因频率分布为4.9%,26.2%,47.6%,16.4%,4.9%。经统计学软件分析显示四川省汉族群体总的基因多样性(GD)值为0.6811。结论研究结果证实四川省汉族群体内部呈现非常显著的遗传异质性,探讨中国不同人群间遗传变异的分布特征和追溯各人群间遗传进化的关系奠定基础。
Objective To study the genetic variation at DYS19 loci on the nonrecombining portion of the Y chromosome in 61 unrelated males from Sichuan H an in China. Methods Blood samples were taken from 61 unrelated males from Sic huan Han in China.Genome DNA was extracted by conventional chelex extraction pro cedure. PCR Fragment analysis was conducted on an ABI PrismTM 310 Genetic Analyz er. Results In all the individuals studied for the DYS19, five alleles were ob tained.These allele frequencyes are 4.9%?26.2%?47.6%?16.4% and 4.9% respectively .Here we obtained gene diversity (GD) for the complete allele was 0.6811 in Sich uan Han in China. Conclusion A larger genetic difference became apparent withi n Sichuan Han. Our foundings display the genetic structural heterogeneity of Ch inese Han populatons and make a significant contribution for tracing genetic evo lutionary relation from Chinese populations.
